Overview

Glucosyltransferase-SI (GtfC) is a critical exoenzyme produced by Streptococcus mutans, the primary bacterial agent associated with dental caries [1, 12]. It belongs to the glycoside hydrolase family 70 and is responsible for the synthesis of both water-soluble and water-insoluble glucans from dietary sucrose [5, 10]. These extracellular polysaccharides form the structural matrix of dental plaque, facilitating the firm attachment of S. mutans and other microorganisms to the tooth enamel [4, 12]. GtfC is particularly notable for its high affinity for the salivary pellicle, making it essential for the initial stages of tooth colonization [4, 5]. As a non-essential enzyme for bacterial survival but a vital factor for virulence, GtfC is a prominent target for anti-caries therapies [1, 3]. Inhibiting GtfC activity reduces the formation of cariogenic biofilms without necessarily killing the bacteria, thereby offering a strategy to prevent tooth decay while minimizing the risk of antibiotic resistance and preserving the commensal oral flora [1, 7].
