Glutamate receptor ionotropic kainate (KAR) represents a distinct family of ligand-gated ion channels that mediate fast excitatory neurotransmission in the central nervous system (Lerma & Marques, 2013). These receptors are composed of tetrameric combinations of five subunits (GluK1–GluK5) and are widely distributed throughout the brain, particularly in the hippocampus and cortex (Contractor et al., 2011). Unlike AMPA receptors, KARs exhibit unique signaling properties, functioning both as ion channels and through G-protein-coupled pathways to modulate synaptic plasticity and neurotransmitter release (Jane et al., 2009). They play a critical role in regulating the balance between excitation and inhibition, and their dysfunction is strongly linked to neurological disorders such as epilepsy, neuropathic pain, and schizophrenia (Lerma & Marques, 2013). Pharmacological targeting of KARs, particularly with subunit-selective antagonists like those targeting GluK1, has shown promise in treating migraines and chronic pain (Contractor et al., 2011). However, the development of therapeutic agents is complicated by the need for high selectivity to avoid off-target effects like sedation or cognitive impairment (Jane et al., 2009).
