Overview

The Glutamate receptor 4 (GluA4), encoded by the GRIA4 gene, is a subunit of the α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptor, which is a ligand-gated ion channel responsible for the majority of fast excitatory neurotransmission in the central nervous system (UniProt P48058). GluA4 is particularly significant during early brain development, where it facilitates the maturation of excitatory synapses and the conversion of silent synapses into functional ones (PubMed: 22114271). In the adult brain, GluA4-containing receptors are characterized by rapid activation and desensitization kinetics, playing specialized roles in high-frequency signaling within circuits like the thalamus and cerebellum. Genetic variations or dysregulation of GluA4 are associated with neurodevelopmental disorders, intellectual disability, and various forms of epilepsy, including absence seizures (NCBI Gene: 2893). Pharmacologically, this subunit is a target for non-competitive antagonists such as perampanel, which are used clinically to manage seizures by dampening overactive glutamatergic signaling (PubMed: 22569160). Additionally, positive allosteric modulators (AMPAkines) are being researched for their potential to improve cognitive deficits in neuropsychiatric conditions by enhancing the duration of the receptor's open state.
