The NMDA receptor is an ionotropic glutamate receptor that plays a critical role in excitatory neurotransmission, synaptic plasticity, learning, and memory. It is a heterotetramer composed of GluN1 and GluN2/GluN3 subunits, and its activation requires the binding of both glutamate and glycine. The receptor is a nonselective cation channel permeable to Na+, K+, and Ca²⁺, and is uniquely blocked by Mg²⁺ ions in a voltage-dependent manner. Dysregulation of NMDA receptor activity is implicated in various neurological disorders.
