Glutamate-rich protein 2 (ERICH2) is a protein encoded by the ERICH2 gene, expressed almost exclusively in the testes in both humans and mice, with minor expression in a few other tissues such as the choroid plexus in developing fetuses and weakly in adult lung and female tissues. The protein is found in the nucleoli fibrillar center and vesicles within testicular cells. Structurally, it contains motifs possibly related to phosphorylation and amidation, and a domain (PHA03247) of uncertain function. Its conserved interactions are mainly with histone H2A family members—including H2AFY, which is involved in X chromosome inactivation—suggesting a role in chromatin dynamics and possibly histone modification. It also interacts with proteins like SDCB1 (involved in vesicle trafficking and cancer cell regulation) and IWS1 (a transcription factor contributing to histone modification and RNA splicing). Despite being highly expressed in the testis, knockout studies in mice reveal ERICH2 is not required for male fertility or reproductive organ development, indicating its function is nonessential in vivo for fertility. There is currently no evidence supporting its role as a therapeutic target, nor are there established drugs, mechanisms of action, or biomarker roles for ERICH2.
