Overview

Glutamate-rich protein 3 (ERICH3) is a large (1,530 amino acid) cytosolic protein with two uncharacterized domains (DUF4590 and DUF4543)[1][2][4]. ERICH3 localizes principally to the primary cilium in diverse cell types and is required for normal cilium formation and length; its loss results in short cilia and abnormal accumulation of transport proteins at the ciliary tip[1][2][5]. ERICH3 is important for retrograde IFT, helping remove signaling proteins (including components of the sonic hedgehog pathway) from cilia. In neurons, ERICH3 is found in the cytoplasm with partial association to vesicular structures, and it interacts with several vesicular trafficking proteins[3][4]. There is evidence of its involvement in regulating plasma serotonin levels and potential antidepressant response, though no specific drugs are currently known to target it[3][4]. Its role in human disease is not well understood, and it is not classified as a canonical therapeutic target such as a receptor, enzyme, transporter, or channel[1][7][5].
