Glutamate-synthesizing enzymes represent a functional group of proteins responsible for the production of glutamate, the primary excitatory neurotransmitter in the central nervous system and a critical intermediate in cellular metabolism. The most prominent members of this group include Glutaminase (GLS), which converts glutamine to glutamate, and Glutamate Dehydrogenase (GDH), which catalyzes the reversible conversion of alpha-ketoglutarate into glutamate. These enzymes play a dual role: in the brain, they regulate the glutamate-glutamine cycle essential for synaptic transmission, while in peripheral tissues and tumors, they support the high metabolic demands of rapidly proliferating cells through anaplerosis. Dysregulation of these enzymes is linked to various pathologies, including neurodegenerative diseases like Alzheimer's and various cancers that exhibit 'glutamine addiction.' Consequently, these enzymes, particularly Glutaminase 1 (GLS1), have become significant therapeutic targets, with small-molecule inhibitors being evaluated for their ability to starve cancer cells of essential nutrients or modulate excitotoxicity in neurological conditions.
