Overview

Glutaminase 2 (GLS2) mRNA encodes a mitochondrial enzyme that catalyzes the hydrolysis of L-glutamine to L-glutamate and ammonia, a process central to hepatic nitrogen metabolism and the urea cycle (UniProt P50136). Beyond its metabolic role, GLS2 is a key mediator of the p53-dependent antioxidant response, facilitating the production of alpha-ketoglutarate and glutathione to combat reactive oxygen species (PubMed: 20385769). In various malignancies, such as hepatocellular carcinoma and breast cancer, GLS2 expression is frequently dysregulated, contributing to metabolic reprogramming and providing a survival advantage to tumor cells under oxidative stress (PubMed: 24658273). Targeting GLS2 mRNA using RNA interference (siRNA) or antisense oligonucleotides (ASOs) represents a therapeutic strategy to disrupt these metabolic dependencies and sensitize cancer cells to apoptosis (PubMed: 30217989). While experimental models have shown that silencing GLS2 mRNA can effectively inhibit tumor growth, clinical development must address potential safety concerns related to systemic ammonia levels and liver function.
