Overview

Glutaredoxin 3 (GLRX3) is a multidomain oxidoreductase enzyme that functions primarily in the cytosol. It binds glutathione and catalyzes GSH-dependent reduction of protein disulfides. GLRX3 is critical for cytosolic iron–sulfur cluster assembly, working as a chaperone for [2Fe–2S] clusters and facilitating maturation of [4Fe–4S] proteins by transferring clusters to essential proteins involved in iron–sulfur assembly[1][3][5]. It regulates cellular redox homeostasis, protects cells from oxidative stress, inhibits apoptosis, and modulates cell survival, often localizing to the nucleus under oxidizing conditions to counteract environmental insults[3][4]. GLRX3 interacts with protein kinase C theta and is implicated in cardiac physiology by regulating hypertrophy and function[4]. Its expression and function have important roles in cancer, iron metabolism, and mitochondrial disorders[4][3].
