Glutathione synthesis refers to the cellular process that produces glutathione (GSH), a tripeptide composed of glutamate, cysteine, and glycine. GSH is a critical intracellular antioxidant and plays essential roles in detoxification, redox homeostasis, maintenance of thiol status, and modulation of cell proliferation. It involves two ATP-dependent enzymatic steps: formation of γ-glutamylcysteine by glutamate–cysteine ligase (GCL), and formation of glutathione by glutathione synthetase (GS). Regulation is primarily through cysteine availability, GCL activity, and cellular demand. Dysregulation or deficiency contributes to various diseases, making manipulation of glutathione synthesis a potential therapeutic strategy.
