The glutathione synthesis pathway is a highly conserved, ATP-dependent biosynthetic process responsible for producing glutathione (GSH), a tripeptide composed of glutamate, cysteine, and glycine. GSH plays critical roles in cellular detoxification, antioxidant defense, maintenance of redox status, and modulation of cell proliferation. The pathway involves two sequential enzymatic steps: the formation of γ-glutamylcysteine (catalyzed by glutamate–cysteine ligase) and the formation of glutathione (catalyzed by glutathione synthetase). Regulation occurs through cysteine availability and feedback inhibition by intracellular GSH. Altered GSH levels are associated with numerous diseases.
