Overview

Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) is a highly conserved, multifunctional enzyme best known for its central role in glycolysis. It catalyzes the sixth step of glycolysis, converting glyceraldehyde-3-phosphate (G3P) into 1,3-bisphosphoglycerate with the concomitant reduction of NAD+ to NADH. While GAPDH’s primary function is in glycolysis, it also has several non-glycolytic roles, including regulation of cell death and survival pathways, participation in nuclear functions such as DNA repair, and interaction with other proteins affecting cellular fate under stress conditions. Altered expression or activity of GAPDH has been implicated in various diseases, including cancer, neurodegenerative diseases, and diabetes.
