Glyceraldehyde 3-phosphate dehydrogenase pseudogene 44 (GAPDHP44) is a processed pseudogene located on chromosome 12q14.2, originating from retrotransposition of the functional GAPDH gene[1][4]. Like other GAPDH pseudogenes, it is generally non-coding, but there is evidence that GAPDHP44 is expressed in the brain and may exert a regulatory role. This pseudogene may influence the risk of late-onset Alzheimer’s disease, as a specific SNP (rs2029721) in GAPDHP44 has been linked to altered disease risk, potentially through mechanisms that involve antisense regulation of the parental GAPDH mRNA or a functional peptide that interacts with proteins such as GOSPEL, affecting cellular stress responses[1]. Unlike the canonical GAPDH enzyme, GAPDHP44 is not a functional enzyme or traditional therapeutic target such as a receptor or transporter, and it is not associated with any approved interacting drugs[1][4][10].
