Overview

Glyceraldehyde 3-phosphate dehydrogenase pseudogene 46 (GAPDHP46) is a genomic sequence highly homologous to the functional enzyme glyceraldehyde-3-phosphate dehydrogenase (GAPDH), but it does not encode a functional protein and lacks biological activity characteristic of GAPDH[6][8]. GAPDHP46 is one of many GAPDH-related pseudogenes present in the human genome[2][8]. These pseudogenes are generally unexpressed and are not implicated in normal physiology or disease pathogenesis[6][8]. The functional GAPDH protein is a classic glycolytic enzyme, but GAPDHP46 itself does not contribute to metabolic or regulatory pathways[6][8]. Pseudogenes such as GAPDHP46 arise from duplication or retrotransposition events, resulting in DNA sequences that are similar to active genes but typically not transcribed or translated into protein[4][8]. Human and mouse genomes contain dozens to hundreds of GAPDH pseudogenes[2][8]. Functional confusion may arise in qPCR/RT-PCR experiments if primers are not specific enough, as GAPDH pseudogenes can be misamplified[8].
