Overview

The glycine binding site is a critical modulatory site on the NMDA receptor, an ionotropic glutamate receptor. It is located on the GluN1 subunit and requires glycine (or D-serine) binding for receptor activation in conjunction with glutamate. Activation of the NMDA receptor allows Ca²⁺ influx, which triggers downstream signaling essential for synaptic plasticity, learning, memory formation, and neurodevelopment. Dysregulation at this site has been implicated in neurological disorders such as schizophrenia and neurodegenerative diseases.

Mechanism of action

Agonist: Binds to the glycine binding site on the GluN1 subunit, enabling NMDA receptor activation upon glutamate binding and subsequent calcium influx. Antagonist: Competitively inhibits glycine binding, preventing NMDA receptor activation.
