The glycine conjugation pathway is a mitochondrial, two-step enzymatic process responsible for the metabolism and detoxification of various endogenous and exogenous compounds. It plays a crucial role in converting potentially toxic substances into more water-soluble, less harmful products that can be excreted in urine. Key enzymes involved are ACSM2B (acyl-CoA synthetase) and GLYAT (glycine N-acyltransferase). Substrates like benzoate and salicylate are conjugated with glycine to form hippuric acid and salicyluric acid, respectively. The pathway maintains CoASH homeostasis, prevents accumulation of toxic organic acids, and influences mitochondrial ATP production. Variations in this pathway may impact susceptibility to liver cancer, musculoskeletal development issues, and metabolic disorders.
