Glycogen synthase 1 (GYS1) is the enzyme responsible for catalyzing the addition of glucose monomers from UDP-glucose to a growing glycogen molecule, forming α-1,4-glycosidic linkages. It plays a critical role in glycogen synthesis and energy storage, primarily in muscle tissue but also in other tissues outside the liver. GYS1 is regulated by phosphorylation (inactivation) and allosteric activation by glucose-6-phosphate. It is a therapeutic target for cancer and metabolic diseases.
