Target intelligence / Profile preview
Glycogen synthase kinase-3 alpha and beta (GSK3α and GSK3β) are closely related serine/threonine kinases that share high amino acid homology in the catalytic domain and are encoded by separate genes (GSK3A, GSK3B)[1][3][5]. They regulate a wide variety of cellular processes by phosphorylating over 100 known protein substrates, participating in diverse signaling pathways such as Wnt/β-catenin, insulin, Notch, and G protein-coupled receptor pathways[1][4][5]. GSK3 activity is tightly regulated by phosphorylation, enabling it to integrate signals from growth factors, hormones, and cellular stress[1][3]. They have fundamental roles in glucose metabolism through regulation of glycogen synthase, but also influence cell growth, differentiation, apoptosis, and neurodevelopment[1][2][5]. Dysregulation of GSK3α/β signaling is implicated in the pathogenesis of cancer, metabolic disorders, and several major psychiatric and neurodegenerative diseases, making these kinases attractive but challenging therapeutic targets[1][3][4][5]. The broad involvement in critical signaling and metabolic processes necessitates careful therapeutic modulation to avoid unintended systemic effects.
Inhibition of GSK3-mediated phosphorylation (e.g., by lithium or small molecule kinase inhibitors); Modulation of downstream pathway activity (e.g., Wnt/β-catenin pathway, insulin signaling).
