Overview

Glycogen synthase kinase-3 alpha and beta (GSK3α and GSK3β) are closely related serine/threonine kinases that share high amino acid homology in the catalytic domain and are encoded by separate genes (GSK3A, GSK3B)[1][3][5]. They regulate a wide variety of cellular processes by phosphorylating over 100 known protein substrates, participating in diverse signaling pathways such as Wnt/β-catenin, insulin, Notch, and G protein-coupled receptor pathways[1][4][5]. GSK3 activity is tightly regulated by phosphorylation, enabling it to integrate signals from growth factors, hormones, and cellular stress[1][3].
