Glycogen synthase kinase-3 beta (GSK-3β) is a multifunctional serine/threonine protein kinase involved in numerous cellular processes, including cell cycle regulation, inflammation, proliferation, differentiation, apoptosis, and neuronal development. It regulates glycogen synthesis, energy metabolism, and cell signaling pathways such as the Wnt/β-catenin pathway. GSK-3β's activity is regulated by phosphorylation at Ser9 (inhibition) and Tyr216 (activation). Dysregulation of GSK-3β is implicated in various diseases, including bipolar disorder, diabetes, neurodegenerative diseases, cancer, and inflammatory disorders, making it a therapeutic target.
Phosphorylation of serine or threonine residues on target proteins, modulating activity. Small molecule inhibitors bind to the ATP binding site, preventing phosphorylation and downstream signaling.
