Glycolipid transfer protein domain-containing protein 2 (GLTPD2) is a 291-residue, cytoplasmic protein coded by the GLTPD2 gene on chromosome 17 and characterized by a C-terminal GLTP-fold domain, a signature of the glycolipid transfer protein (GLTP) superfamily[1][2]. It is predicted to facilitate the binding and intermembrane transfer of lipids—particularly ceramide 1-phosphate and glycolipids—playing a role in sphingolipid transport and homeostasis, though its tissue distribution and precise physiological roles remain only partially characterized[2][4]. Structural analyses suggest the presence of an N-terminal transmembrane-like helix, likely stabilizing membrane interactions and functioning near membrane contact sites[2]. GLTPD2 shares homology with ceramide 1-phosphate transfer protein (CPTP) and has no reported paralogs outside the GLTP superfamily[2][4]. While other GLTP family proteins are implicated in autophagy, inflammasome activation, cytokine secretion, and programmed cell death, direct evidence for such roles specifically for GLTPD2 is still emerging and largely unexplored as of 2025[2].
