Overview

Glycolysis-associated long non-coding RNA of thyroid cancer (GLTC), also known as GLTC1 or lnc-UNC93B1-1, is a long non-coding RNA upregulated in papillary thyroid cancer (PTC). GLTC acts as an oncogenic regulator by directly interacting with lactate dehydrogenase A (LDHA) and promoting its succinylation at lysine 155. This enhances LDHA enzymatic activity and increases aerobic glycolysis—commonly known as the Warburg effect—which supports tumor cell proliferation, growth, and distant metastasis. High GLTC expression correlates with poor prognosis and radioiodine resistance in PTC. Because of its clear mechanistic role and clinical association, GLTC is considered a promising therapeutic target and prognostic biomarker for papillary thyroid cancer[2][4].

Mechanism of action

Promotes succinylation of LDHA at K155 by inhibiting SIRT5-LDHA interaction, enhancing LDHA enzymatic activity and glycolysis, induces radioiodine resistance in thyroid cancer cells[2].
