gp41 is a transmembrane glycoprotein that forms part of the envelope spike complex of Human Immunodeficiency Virus type 1 (HIV-1). It plays a critical role in mediating the fusion between the viral and host cell membranes, enabling viral entry into target cells. Encoded by the env gene as part of gp160, gp41 contains several distinct regions including the fusion peptide, heptad repeats (HR1/NHR and HR2/CHR), the membrane proximal external region (MPER), and a transmembrane domain. After HIV binds to CD4 receptors via gp120, conformational changes expose/activate gp41 facilitating membrane fusion. It is a key target for entry/fusion inhibitor drugs and vaccine design.
