Overview

Glycoprotein carbohydrate moieties, commonly referred to as glycans, are complex sugar chains covalently attached to proteins through N-linked or O-linked glycosylation. These moieties play critical roles in biological processes including protein folding, stability, and cell-to-cell communication by serving as ligands for various receptors (Varki et al., 2022, Essentials of Glycobiology). In many diseases, particularly cancer, glycosylation patterns become aberrant, leading to the expression of tumor-associated carbohydrate antigens (TACAs) that promote metastasis and immune evasion (Pinho & Reis, 2015, Nature Reviews Cancer). Therapeutic strategies targeting these moieties include the use of monoclonal antibodies against specific glycan epitopes, small molecule inhibitors of glycosylation enzymes, and glycan-mimetics designed to disrupt pathogen binding or inflammatory signaling (Munkley & Elliott, 2016, International Journal of Molecular Sciences).
