Target intelligence / Profile preview

Glycoprotein non-metastatic melanoma protein B (GPNMB)

Target
GPNMB
Molecular classification
Type I transmembrane glycoprotein
01

Overview

Glycoprotein NMB (GPNMB) is a type I transmembrane glycoprotein involved in cell adhesion, migration, differentiation, and bone remodeling. It is overexpressed in several cancers and plays a role in immune modulation and inflammation. As a therapeutic target, GPNMB is being explored for cancer immunotherapy using antibody-drug conjugates. It is also implicated in cardiovascular repair following myocardial infarction, suggesting potential therapeutic applications in regenerative medicine.

Mechanism of action

Glembatumumab vedotin/CDX-011: Antibody-drug conjugate that binds GPNMB, internalizes into the cell, and releases a cytotoxic payload (vedotin) to kill cancer cells.
