Glycosaminoglycan (GAG) biosynthesis is the cellular process of assembling GAGs, linear polysaccharides, and attaching them to core proteins to form proteoglycans. GAGs are essential components of the extracellular matrix and cell surfaces, crucial for cell signaling, structural integrity, and tissue hydration. The biosynthesis involves activated sugar donors, formation of a tetrasaccharide linker, polymerization, and modification reactions such as sulfation and epimerization. Key enzymes include xylosyltransferases (XYLT1/2), galactosyltransferases (B4GALT7, B3GALT6), glucuronosyltransferase (GLCAT1), EXTL3, CSGALNACT1/2, exostosins (EXT1/2), and NDST enzymes. Defects in these enzymes can lead to connective tissue disorders.
