Overview

Glycosaminoglycan regulatory associated long non-coding RNA (GRASLND), also known as RNF144A-AS1, is a primate-specific long non-coding RNA (lncRNA) regulator that plays a critical role in mesenchymal stem cell chondrogenesis and cartilage matrix production, primarily via suppression of the interferon-gamma signaling pathway through EIF2AK2 binding[1]. GRASLND is a key regulator of cartilage differentiation and has therapeutic potential for cartilage regeneration and osteoarthritis treatment[1]. Furthermore, it is overexpressed in various cancers, including melanoma, bladder, gastric, and ovarian cancer, where it promotes tumor proliferation, invasion, metastasis, angiogenesis, and immune evasion—most notably by suppressing IFN-γ-stimulated anti-tumor responses[2][3][4][5][6]. GRASLND is considered an unfavourable prognostic marker in several cancer types, and modulating its expression may offer diagnostic and therapeutic opportunities across oncology and regenerative medicine.

Mechanism of action

Therapeutic manipulation (overexpression, silencing, CRISPR-dCas9 activation) to regulate stem cell chondrogenesis and cartilage regeneration. Suppression of IFN-γ signaling pathway by direct interaction with EIF2AK2. Promotion of tumorigenic signaling pathways such as YAP1. Regulation of EMT via interactions with miRNAs and key proteins (e.g., LOX).
