Glycosyltransferase 8 domain-containing protein 1 (GLT8D1) is a human enzyme of the glycosyltransferase family, specifically the GT8 family, encoded by the GLT8D1 gene[3][7][8]. It is a type II membrane glycoprotein localized to the Golgi and endoplasmic reticulum, with a short cytoplasmic domain, a transmembrane region, and a large lumenal catalytic domain[1]. GLT8D1 is involved in the transfer of glycosyl residues from sugar nucleotide donors to acceptor molecules, contributing to the formation of glycosidic bonds, though its precise endogenous substrates and full enzymatic activities remain incompletely defined[1][3]. Structurally, it contains three N-glycosylation sites and is closely related to other CAZy GT8 family members. Mutations in GLT8D1 are associated with familial amyotrophic lateral sclerosis, and its overexpression in cancers, such as gastric cancer and glioma, promotes proliferation, cell cycle progression, and resistance to apoptosis, partly through modulation of the JAK2/STAT3 signaling pathway[1][2][6]. Elevated GLT8D1 expression is linked to poor prognosis in several cancers, positioning it as a potential biomarker and possible but as yet untested therapeutic target[2][6][7]. No specific drugs are currently identified as interacting with GLT8D1, and direct pharmacological inhibition has not been described in current literature.
