Glycosyltransferase GtfA is a specialized bacterial enzyme primarily recognized for its role in the post-translational modification of serine-rich repeat proteins (SRRPs), which are critical surface adhesins in Gram-positive bacteria such as Streptococcus gordonii and Staphylococcus aureus (UniProt P0C0H5). Working often in a heterodimeric complex with GtfB, GtfA catalyzes the initial steps of glycosylating these large proteins, a process that is essential for their stability, transport to the cell surface, and subsequent ability to bind host receptors like those on human platelets (PubMed 24464062). This adhesion is a fundamental step in the development of life-threatening conditions such as infective endocarditis and various respiratory infections (PubMed 21940797). As an anti-virulence target, GtfA is highly attractive because its inhibition can neutralize a pathogen's ability to colonize and cause disease without necessarily killing the bacteria, thereby reducing the selective pressure for antibiotic resistance. While no drugs targeting GtfA are currently FDA-approved, experimental small-molecule inhibitors, including 2-aminobenzimidazole derivatives, have demonstrated the ability to disrupt SRRP glycosylation and reduce bacterial adhesion in preclinical models (PubMed 27161314). The absence of close human homologs for this specific glycosylation pathway suggests that GtfA inhibitors could offer a high degree of selectivity with minimal off-target toxicity.
