Overview

Glypican-1 (GPC1) mRNA is the messenger RNA transcript that encodes the GPC1 protein, a cell surface heparan sulfate proteoglycan (HSPG) attached to the membrane via a glycosylphosphatidylinositol (GPI) anchor. This molecule serves as a critical template for the production of GPC1, which acts as a co-receptor for various heparin-binding growth factors, including fibroblast growth factors (FGFs) and vascular endothelial growth factor (VEGF), thereby regulating cell signaling, proliferation, and angiogenesis (PubMed: 9537005). GPC1 mRNA is frequently overexpressed in several aggressive cancers, most notably pancreatic ductal adenocarcinoma and glioblastoma, where it promotes tumor growth and metastasis (PubMed: 16452190). As a therapeutic target, GPC1 mRNA is approached using RNA interference (RNAi) or antisense oligonucleotides (ASOs) to silence the expression of the GPC1 protein, effectively disrupting the oncogenic signaling pathways it supports (PubMed: 28213458). Furthermore, the detection of GPC1 mRNA and its protein product in circulating exosomes has emerged as a promising non-invasive biomarker for early cancer detection and monitoring (Nature: 26108854). Therapeutic challenges include the efficient delivery of nucleic acid-based drugs to the tumor site and avoiding off-target effects within the transcriptome.
