Overview

Glypican 3 antisense transcript 1 (GPC3-AS1) is a long noncoding RNA (lncRNA) transcribed antisense to the glypican 3 (GPC3) gene. Highly upregulated in hepatocellular carcinoma (HCC) tissues, GPC3-AS1 acts as an oncogenic RNA that promotes tumor growth, proliferation, and migration both in vitro and in vivo. Mechanistically, GPC3-AS1 functions as an epigenetic activator by recruiting the histone acetyltransferase P300/CBP-associated factor to the GPC3 gene body, inducing histone modifications associated with active chromatin and thereby increasing GPC3 expression. The expression of GPC3-AS1 correlates with clinically adverse features in HCC, such as larger tumor size and worse prognosis, suggesting its potential as a diagnostic and prognostic biomarker. GPC3-AS1 is under investigation as a potential therapeutic target due to its role in the epigenetic activation of oncogenic pathways in liver cancer[1][3].
