Overview

Glypican 5 antisense RNA 1 (GPC5-AS1) is a long non-coding RNA transcribed from the antisense strand relative to the GPC5 gene on chromosome 13q31.3. GPC5-AS1 functions as an endogenous molecular sponge for microRNAs miR-93 and miR-106a, protecting GPC5 mRNA from degradation and thereby enhancing the expression of this putative tumor suppressor. In gastric cancer, GPC5-AS1 is frequently downregulated, contributing to increased cell proliferation, impaired apoptosis, and accelerated tumor progression. Overexpression of GPC5-AS1 can suppress gastric cancer cell growth, induce apoptosis, and retard tumorigenesis in vitro and in vivo. Epigenetically, the promoter activity of GPC5-AS1 is regulated by methyl-CpG-binding protein 2 (MeCP2), affecting its expression in cancer cells. Studies suggest GPC5-AS1 may serve as both a diagnostic biomarker and a promising therapeutic target for gastric cancer. Its roles in other cancer types are under bioinformatic investigation, though experimental evidence is limited beyond gastric cancer[1][2][3][4].
