The goblet cell mucin secretion pathway encompasses the cellular and molecular mechanisms by which goblet cells, specialized epithelial cells in the respiratory and gastrointestinal tracts, synthesize, store, and secrete mucins to form a protective mucus gel layer. Mucin secretion occurs through basal (constitutive) and regulated pathways, the latter often involving purinergic (P2Y) receptor signaling, calcium-dependent exocytosis (involving SNARE and synaptotagmin proteins), and is stimulated by environmental irritants, pathogens, and inflammatory mediators. While essential for tissue protection and immune modulation, dysregulation of these pathways—resulting in over- or under-secretion of mucin—co ntributes to disease processes such as asthma, chronic obstructive pulmonary disease, cystic fibrosis, and inflammatory bowel disease.
