Golgi alpha-mannosidase II (GMII) is a key glycosyl hydrolase enzyme involved in the N-linked glycosylation pathway. It catalyzes the removal of two mannosyl linkages from GlcNAcMan\(_5\)GlcNAc\(_2\), a committed step in complex N-glycan synthesis. GMII is essential for the maturation of N-glycans on glycoproteins within the Golgi apparatus. Inhibition of GMII has shown promise as a therapeutic strategy against certain cancers. Mutations in the MAN2A1 gene, encoding GMII, can disrupt normal glycoprotein processing and, in mice, lead to systemic autoimmune disease phenotypes.
