Overview

Golgi-associated olfactory signaling regulator (GFY), also known as Goofy, is an integral membrane protein primarily localized to the Golgi apparatus of olfactory and vomeronasal sensory neurons[6][5][3]. GFY is specifically expressed in the olfactory epithelium, where it is required for normal acuity of olfactory signaling[2][4][6]. Functional studies in mice show that deletion of GFY (Goofy) impairs odor sensitivity due to the mislocalization of adenylyl cyclase III, stunted olfactory cilia, and weakened physiological and behavioral responses to odorants[6][2][4]. GFY appears to regulate cilia development in olfactory neurons and the trafficking of key olfactory signal transduction proteins, thus affecting overall olfactory sensitivity[6][2][3]. Although its physiological role in humans is not fully elucidated, GFY is predicted to be involved in human olfactory perception, and its dysfunction may contribute to anosmia or other disorders of smell[2][4]. GFY is **not currently classified as a therapeutic receptor, enzyme, transporter, or drug target.**
