Golgi-associated RAB2 interactor family member 3 (GARIN3) is a protein-coding gene member of the GARIN family, which act as effectors for RAB2A and RAB2B small GTPases that regulate membrane trafficking processes[1][3]. GARIN3 contains a RAB2-binding domain and is recruited to the Golgi apparatus by RAB2A/B in cells[1]. It is widely expressed but has notable functional importance during spermiogenesis, especially in sperm head morphogenesis and acrosome biogenesis. Knockout studies in mice show that GARIN3 is essential for male fertility; deletion leads to abnormal sperm head shape, impaired acrosome formation, and reduced levels of several sperm head plasma membrane proteins (such as ADAM2, ADAM3, ADAM32), as well as significant male infertility[1]. Unlike GARIN1B, GARIN3 can bind both the active (GTP-bound) and inactive (GDP-bound) forms of RAB2A/B, suggesting functional diversity within the GARIN family[1]. The protein may also be involved in RNA biogenesis, but this function remains predicted and not experimentally confirmed[3]. There are currently no known drugs, biomarkers, or specific safety concerns associated with GARIN3, and it is not considered a therapeutic target.
