Golgi-associated RAB2 interactor family member 5B (GARIN5B) is a Golgi-associated protein primarily expressed in testes, where it plays an essential role in the morphogenesis of the sperm head during spermiogenesis. The protein contains a RAB2-binding domain, conferring affinity for the small GTPases RAB2A and RAB2B, which mediate Golgi-to-acrosome trafficking processes. Knockout mouse models show that GARIN5B is indispensable for sperm morphological integrity and, when deleted, lead to subfertility due to aberrant sperm development. Its close paralogs include GARIN3, and the GARIN protein family as a whole is conserved in humans and mice. Currently, GARIN5B is not a recognized therapeutic target in clinical medicine, nor are there any drugs or established biomarkers related to its activity.
