Golgi phosphoprotein 3-like (GOLPH3L) is a paralogue of Golgi phosphoprotein 3 (GOLPH3), found in vertebrate genomes. It binds to phosphatidylinositol 4-phosphate (PI4P), localizes to the Golgi apparatus in cells, and is required for efficient protein trafficking in secretory cells. Unlike GOLPH3, which interacts tightly with myosin 18A (MYO18A) and is an oncogene, GOLPH3L doesn't bind MYO18A and acts as an antagonist to GOLPH3, modulating Golgi morphology by promoting compaction when overexpressed and dispersal when depleted. Its expression is restricted to tissues with high secretory activity, such as salivary glands and small intestine, and it is thought to play a role in maintaining optimal secretion rates by counterbalancing GOLPH3's effects on the Golgi apparatus. GOLPH3L is not a receptor, enzyme, or transporter, but rather a regulatory phosphoprotein of the Golgi apparatus. Its functional role is to antagonize GOLPH3, affecting Golgi structure and trafficking, most evident in secretory tissues. There is currently no therapeutic or diagnostic application for GOLPH3L, nor is it directly implicated in disease.
