Gonadotropin-releasing hormone 1 (GNRH1) is a decapeptide neurohormone synthesized in the hypothalamus and released in a pulsatile manner into the hypophyseal portal system. It stimulates the anterior pituitary to secrete luteinizing hormone (LH) and follicle-stimulating hormone (FSH), which in turn regulate gonadal steroidogenesis and gametogenesis. GNRH1 is the central regulator of the hypothalamic-pituitary-gonadal axis in vertebrates. Clinically, GNRH1 and its synthetic analogs serve as key therapeutic agents for reproductive disorders, hormone-dependent cancers, and as tools for pituitary function testing. Overexpression or aberrant signaling of GnRH or its receptor is implicated in various hormone-dependent malignancies, establishing it as an important therapeutic target.
Agonists stimulate pituitary GnRH receptors to cause FSH and LH release; antagonists block these receptors to suppress gonadotropin secretion; both approaches modulate sex steroid production for clinical use in hormone-sensitive cancers and reproductive disorders
