The putative gonadotropin-releasing hormone II receptor (GnRHR2) is a predicted G protein-coupled receptor believed to bind gonadotropin-releasing hormone II (GnRH-II). In most mammals, including humans, the GNRHR2 gene exists but is non-functional due to frameshift mutations and premature stop codons. This means that while a gene homolog for the receptor is present in the human genome, it does not encode a full-length or functionally expressed protein. There is some limited evidence from certain species (such as pigs and some non-human primates) that functional variants of this receptor exist, but in humans, the receptor is considered putative and most likely non-functional. Functional GnRH-II receptors have been described in some non-human species, where they regulate reproductive and other peripheral functions, but this does not apply to humans. Therefore, in humans, the GnRHR2 gene is present, but a functional, expressed protein is not produced. It is not currently considered a drug target in humans, and no approved drugs or experimental compounds are known to interact specifically with a human functional GnRHR2, nor are there drugs or biomarkers directly associated with this protein in humans. The classification as a receptor and GPCR is based on sequence homology and experimental data from non-human species, not functional protein expression in humans.
