Overview

The gonadotropin-releasing hormone (GnRH) pathway is a central regulator of reproductive function. GnRH, produced in the hypothalamus, stimulates the anterior pituitary to release LH and FSH. These gonadotropins are essential for sexual development, fertility, and regulation of the menstrual cycle in females and spermatogenesis in males. The pathway is regulated by feedback from sex steroids and other factors. Dysregulation is associated with various reproductive disorders, and the pathway is a major target for infertility treatments and therapies aimed at reducing sex steroid-dependent tissue growths.

Mechanism of action

GnRH agonists stimulate the GnRH receptor initially, but prolonged use leads to desensitization and downregulation. GnRH antagonists directly block the GnRH receptor.
