Overview

GPRC5D antisense RNA 1 (GPRC5D-AS1) is a long non-coding RNA transcribed from the antisense strand of the GPRC5D locus, and also associated with HEBP1[1][3]. It is located on human chromosome 12p13.1. In cancer contexts, particularly renal cell carcinoma, GPRC5D-AS1 functions as a tumor suppressor: downregulation of GPRC5D-AS1 results in increased proliferation, invasion, and migration of cancer cells, possibly by activating the Wnt-β-catenin signaling pathway and epithelial-mesenchymal transition. In sarcopenia, GPRC5D-AS1 regulates muscle cell ferroptosis through direct interaction with and upregulation of SLC7A11, reducing cellular iron-induced lipid peroxidation and cell death. Collectively, GPRC5D-AS1 is emerging as a disease-specific biomarker and potential therapeutic target, though its non-coding RNA nature presents unique targeting and validation challenges[1][3].
