Grainyhead-like transcription factor 1 (GRHL1) is a member of the grainyhead family of transcription factors with a highly conserved, immunoglobulin-like DNA binding domain structurally related to p53[2]. GRHL1 plays a critical role in regulating epidermal differentiation and the expression of cell adhesion molecules, such as desmoglein 1, which is essential for maintaining skin integrity and proper hair anchorage[3][5]. It functions predominantly as a homodimer or may form heterodimers with related family members and is important during embryonic development[2][5]. In cancer biology, GRHL1 has context-dependent roles, acting as a tumor suppressor or promoter depending on tissue type: it is associated with altered cancer cell proliferation, and its dysregulation affects cell cycle genes[1]. Loss of GRHL1 in animal models leads to skin barrier defects, abnormal desmosomes, and increased risk for malignant progression to squamous cell carcinoma[3]. Mutations in the GRHL1 gene are also linked to inherited deafness and corneal dystrophy[5]. As a potential drug target, its involvement in regulating cell cycle genes in cancer is under investigation, but no specific drugs are currently known to target it directly[1][4][5].
