GRAM domain-containing protein 2B (GRAMD2B) is a mammalian protein characterized by a GRAM domain and a transmembrane domain that anchors it to the endoplasmic reticulum (ER). It is structurally related to yeast lipid transfer proteins (the LAM family), though unlike some homologues, GRAMD2B lacks the VASt domain associated with sterol transport. Its biological function has not been clearly defined, but it is presumed to play a role in inter-organelle lipid transfer and the organization of membrane contact sites in cells, based on similarity to its paralogues. To date, there is no reported link between GRAMD2B and human disease, and it is not described as a therapeutic target. Its gene has several alternative spliced transcripts but no evidence of coding for a functionally characterized protein. The current nomenclature clarifies past confusion with aliases such as GRAMD3 and NS3TP2, designating GRAMD2B as the recommended canonical name.
