GRAM domain-containing protein 4 (GRAMD4) is a mitochondrial adaptor protein that mediates apoptosis independently of p53, primarily under regulation by the transcription factor p73. Following cellular stress or DNA damage (e.g., chemotherapeutic agent cisplatin), p73 activates GRAMD4 transcription. GRAMD4 then translocates to mitochondria, interacts with the anti-apoptotic protein Bcl-2, and promotes the activation and oligomerization of Bax, leading to mitochondrial membrane permeabilization, cytochrome c/Smac release, and caspase-3 activation. The protein contains a GRAM domain, two transmembrane domains, and a Bcl-like region facilitating protein-protein interactions in the apoptotic cascade. It is implicated in the execution phase of programmed cell death and has potential relevance in cancer therapy and biomarker development
Drugs like cisplatin upregulate p73, which directly transactivates GRAMD4. GRAMD4 translocates from nucleus to mitochondria, binds Bcl-2, releases apoptotic factors (cytochrome c, Smac), and induces caspase-3 activation
