Gram-negative outer membrane proteins and other macromolecules encompass a broad array of essential bacterial components that maintain cellular integrity and facilitate survival within host environments. The outer membrane is a unique bilayer containing lipopolysaccharides (LPS) and various outer membrane proteins (OMPs), such as porins and efflux pumps, which control the influx of nutrients and the efflux of toxic substances, including antibiotics (Silhavy et al., 2010, Cold Spring Harb Perspect Biol). Other vital macromolecules include the peptidoglycan cell wall, which provides mechanical strength, and internal structures like ribosomes and nucleic acids that are central to protein synthesis and genetic replication (Wilson, 2014, Nat Rev Microbiol). These components are the primary targets for various classes of antibiotics; for instance, polymyxins disrupt the LPS layer, while beta-lactams inhibit peptidoglycan synthesis (Falagas & Kasiakou, 2005, Clin Infect Dis). Targeting these structures is a cornerstone of treating infections caused by multidrug-resistant Gram-negative pathogens, though challenges such as membrane impermeability and active efflux often limit drug efficacy (Breijyeh et al., 2020, Molecules). This target entry is considered incorrect as a single entity because it aggregates numerous distinct molecular targets into a single descriptive category.
