Overview

Smart-Gram-positive bacterium** refers to a broad class of bacteria that retain the crystal violet stain in the Gram staining procedure due to their thick peptidoglycan cell wall[8][1][5]. This group is defined by structural features such as a cytoplasmic lipid membrane, a thick peptidoglycan layer, and teichoic acids within the cell wall[8]. They lack an outer membrane found in gram-negative bacteria. The classification includes two major groups based on DNA G+C content: high G+C (Actinobacteria) and low G+C (Bacilli)[1][5]. Gram-positive bacteria are not a single molecular target but rather an entire category of organisms. As such, they are not considered a specific therapeutic target like an enzyme or receptor; instead, various molecular components within these bacteria—such as enzymes involved in cell wall synthesis or quorum-sensing systems—are targeted for antimicrobial therapy[7][3]. Common pathogens include Staphylococcus aureus, Streptococcus species, and Enterococcus species. Therapeutics against gram-positive infections often exploit differences in their cell envelope structure. Drugs like vancomycin and beta-lactams inhibit peptidoglycan cross-linking or synthesis[2][9], while newer agents may target other essential bacterial processes. Resistance among gram-positive pathogens is clinically significant; examples include methicillin-resistant Staphylococcus aureus (MRSA) and vancomycin-resistant enterococci (VRE)[2].
