Overview

The granulin precursor protein (progranulin) is a secreted glycoprotein that plays diverse roles in cell growth, wound healing, inflammation, lysosomal function, and neuronal survival. It is the precursor for granulins, a family of smaller peptides generated by proteolytic cleavage. Mutations or deficiencies in progranulin are associated with frontotemporal lobar degeneration (FTLD), neuronal ceroid lipofuscinosis and Lupus nephritis. It acts both extracellularly via signal transduction pathways and intracellularly via transcriptional modulation. It's involved in normal lysosome function, and deficiency impairs cellular waste processing.
