Overview

Granulocyte activity refers to the collective physiological functions and activation states of granulocytes, which include neutrophils, eosinophils, and basophils [5]. These cells represent a critical component of the innate immune system, providing host defense through mechanisms such as chemotaxis, phagocytosis, the release of antimicrobial enzymes via degranulation, and the generation of reactive oxygen species (ROS) during an oxidative burst [3, 9]. While essential for clearing bacterial and fungal infections, aberrant or excessive granulocyte activity is a central pathological feature of chronic inflammatory conditions, autoimmune diseases, and acute tissue damage seen in ischemia-reperfusion injury [5, 6]. In the oncology setting, certain granulocyte subsets can exhibit immunosuppressive properties that facilitate tumor evasion or promote progression through the release of pro-inflammatory cytokines [4]. Consequently, pharmacological strategies often target the pathways regulating these cells—either to stimulate granulocyte counts in patients with neutropenia or to suppress their harmful activation in allergic and autoimmune disorders [4, 6].

Mechanism of action

Therapeutic modulation of granulocyte activity is achieved by agents that either stimulate production and survival through colony-stimulating factor receptors (e.g., G-CSFR, GM-CSFR) or suppress cellular activation, recruitment, and effector functions through glucocorticoid receptors and various anti-inflammatory signaling pathways [4, 6, 7].
