Overview

The granulocyte-macrophage colony-stimulating factor receptor alpha subunit (GM-CSFRα), also known as CSF2RA or CD116, is a critical component of the heterodimeric GM-CSF receptor complex. This receptor mediates the biological effects of granulocyte-macrophage colony-stimulating factor (GM-CSF), a cytokine that regulates the production, differentiation, and function of granulocytes and macrophages. It consists of an alpha chain (GM-CSFRα), responsible for ligand binding, and a beta chain (βc), shared with IL-3 and IL-5 receptors, responsible for signal transduction. GM-CSF first binds to GM-CSFRα with low affinity, followed by recruitment of βc to form a high-affinity complex, leading to activation of downstream signaling pathways like JAK/STAT, MAPK, and PI3K.
