Overview

GRB10 interacting GYF protein 1 (GIGYF1) is an adaptor protein belonging to the GYF family, characterized by a GYF protein interaction domain. GIGYF1 interacts with GRB10, an adaptor protein that modulates signaling downstream of the insulin and IGF-1 receptors. Through these interactions, GIGYF1 regulates receptor signaling, enhances insulin and IGF-1 receptor phosphorylation, and participates in metabolic and growth pathways. Beyond this, GIGYF1 is a translational repressor: it binds the translation initiation machinery (specifically, the eIF3 complex) and disrupts interactions necessary for efficient mRNA translation, notably repressing the production of certain transcripts such as interferon-β. GIGYF1 is broadly expressed and may play diverse roles in translational control, genome stability, and metabolic regulation. Loss-of-function mutations in GIGYF1 are associated with a range of disorders, from neurodevelopmental syndromes (including autism spectrum disorder) to metabolic dysfunction (including diabetes and altered glucose homeostasis). No approved drugs specifically target GIGYF1 at this time.
